چکیده

Abstract Bacterial overgrowth is common for improperly stored urine. However, its effects on human urinary extracellular vesicles (uEVs) study had not been previously examined nor documented. This investigated the presence of bacterial EVs (bEVs) contaminated in uEVs samples and their study. Nanoscale were isolated from normal urine immediately after collection (0‐h) or 25°C‐storage with/without preservative (10 mM NaN 3 ) up to 24‐h. Turbidity, count total proteins abnormally increased 8‐h 24‐h‐stored without . NanoLC‐ESI‐LTQ‐Orbitrap MS/MS identified 6–13 these samples. PCR also detected DNAs Besides, derived 24‐h induced macrophage activation (CD11b phagocytosis) secretion cytokines (IFN‐α, IL‐8, TGF‐β) macrophages renal cells (HEK‐293, HK‐2, MDCK). All by contamination partially/completely prevented Interestingly, cytokine bEVs purified Escherichia coli clearly shows evidence when inappropriately stored, whereas can prevent such bEVs.

EXTRACELLULAR VESICLES AND THEIR THERAPEUTIC APPLICATIONS: A REVIEW

Background & Aims: Extracellular vesicles are double phospholipid layer vesicles releasing from several cells.  These particles contain bio-molecules including mRNA, miRNA, DNA, proteins, and lipids which play a pivotal role in the cell to cell communication. Once delivered, they participate to change target cell function, fate, morphology, differentiation, and growth. The present study is a re...

Isolation and characterization of Faecalibacterium prausnitzii extracellular vesicles

Introduction: Extracellular vesicles (EVs) contain active biological compounds which play important roles in biological processes. The secretion of EVs is a common phenomenon which occurs in archaea, bacteria and mammalian cells. The secretion of bacterial EVs has been discovered in various species of both Gram-negative and Gram-positive bacteria. Faecalibacterium prausnitzii is one of the comm...

Extracellular Vesicles in Regenerative Medicine, a Brief Review

Extracellular vesicles were initially known as cellular waste carriers, while recent studies demonstrate that extracellular vesicles play important biological roles in all aspects of life-from single cells to mammalians. Their pathophysiological roles in some diseases like cancer are being decoded. Extracellular vesicles are divided into some classes and there are different strategies to isolat...

Aquaporins in Urinary Extracellular Vesicles (Exosomes)

Since the successful characterization of urinary extracellular vesicles (uEVs) by Knepper's group in 2004, these vesicles have been a focus of intense basic and translational research worldwide, with the aim of developing novel biomarkers and therapeutics for renal disease. Along with these studies, there is growing evidence that aquaporins (AQPs), water channel proteins, in uEVs have the poten...

Extracellular Vesicles as Neprilysin Delivery Syemory Improvement of Alzheimer’s Disease

Alzheimer’s disease (AD) is a neurodegenerative brain disorder which has no effective treatment yet due to the blood barrier in the brain that limits the drugs with the potential of disease improvement. Extracellular vesicles (EVs) are biocompatible nanoparticles with a lipid membrane. These vesicles are secreted from various cells such as mesenchymal stem cells (MSCs) and can pass through biol...
